- Duplicate samples from the freshly prepared test media of the only test concentration of nominal 100 mg/L and the control were taken at the start of the test.
For the determination of the stability of the test item under the test conditions and of the maintenance of the test item concentrations during the test period, duplicate samples from the test media of the only test concentration of nominal 100 mg/L and the control were collected at the end of the test (after 48 hours) by pouring together the contents of the test beakers of each treatment.

The Daphnia introduced in the test were taken from our in house laboratory culture.
The Daphnia were bred in the laboratories of the tesing facility under similar temperature and light conditions as used in the test. The cultivation of the parental Daphnia was performed in reconstituted water of a similar quality with regards to pH, the constituent salts and total hardness as the test medium used in the test. The test organisms were not first brood progeny. Once a week the Daphnia in the stock culture were fed with Tetra Min-extract and at least all other working days with green algae (Desmodesmus subspicatus) freshly grown in the laboratories of the testing laboratory.

The purpose of this study was to evaluate the influence of the test item Me3K4A2 on the mobility of Daphnia magna according to OECD Guideline 202. Young Daphnia (< 24 hours old) were exposed in a static test for 48 hours to an aqueous test medium containing the test item at the a concentration of 100 mg/l nominal. The test substance is virtually insoluble in water. This limit test was performed in compliance with the test guidelines to demonstrate that the test item has no toxic effect on Daphnia up to at least this concentration.
To avoid physical effects of undissolved test item on Daphnia no concentration above the solubility limit of the test item in test medium was tested. Therefore, a supersaturated stock solution of 100 mg test item/L wa sprepared by suspending 150 mg of test item in 1500 mL of test medium for preparing the test concentration. The stock suspension was stirred for 25.5 hours in the dark to dissolve as much test item as possible. Then, the undissolved test item was separated by filtration and the filtrate was used as test medium. The test concentration was prepared just before introduction of theDaphnia(= start of the test).After 24 hours in the control and in the only test concentration of nominal 100 mg test item/L no immobilization of the test animals was observed.
