- All test medium samples for the analytical determinations were taken from the approximate center of the test vessels ( without mixing the medium ) and kept at -18 °C to -25 °C until analyses.

0.5 g test substance were mixed with 200 ml test medium and filled up to 500 ml with test medium. The stock solution was homogenized for 20 minutes by ultrasonic treatment. Since the test substance was poorly soluble in test medium, most of the test substance settled down and built a sediment at the bottom of the flask.
Test concentrations
Based on a pretest, the nominal test concentrations were chosen as 58, 100 and 180 mg test article /l . For the preparation of the test concentrations only the supernatant of the stock solution was taken. - Test organisms (species):

- The organism used was Daphnia magna. The Daphnia were bred in the laboratories of the Product Ecology TF, Textile Dyes Division under standardised conditions.
Breeding :
Cultures of Daphnia magna are held in glass vessels containing aprox. 3.0 litre of reconstituted water at 18 - 22 °C. The waterwas renewed partially three times per week. The Daphnia were feed with a suspension of green algae (Scenedesmus subspicatus) supplemented by a TETRAMIN-extract in such quantités that the food was consumed within 24h.
Pretreatment :
24 hours before the start of the exposure reproductive Daphnia were separated from the young Daphnia by sieving all individuals through a 1000 µm sieve.
For each concentration, of test substance, 20 Daphnia divided in two groups of 10 animals are used. - Test type:

The short term toxicity of FAT 20042/D to Daphnia magna was determined in a 48-hour static test according to the OECD Guideline 202, adopted 4 April 84, and EEC Guideline 84/449, L 251, Part C 2. The test concentrations were prepared from 1.0 g/l stock solution, in which most of the test substance settled down. For the preparation of the test concentrations only the supernatant of the stock solution was taken ( limit of the solubility ). The nominal concentrations tested were 58, 100 and 180 mg test article/l, and a control without any additions. Because of the low solubility of the test article in the test media the actual test concentrations in the analysed test media was in the range of 7.7 - 12.2 % of the nominal values. Therefore, the results were reported related to the avarage analytical concentrations of the test article measured at the beginning and at the end of the test, which were determined to be 6.1, 8.9 and 14.3 mg/L for nominal concentrations of 58, 100 and 180 mg/L respectively. No immobilisation and abnormal behaviour of the Daphnia was observed in the control and in all test concentrations. Hence, the NOEC and EC0 value were determined to be 14.3 mg/l, while the EC50 and the EC100 values are clearly higher than 14.3 mg/l after 48 hours exposure.
