Stock solutions (150 and 75.0 mg/L) were freshly prepared with dilution water. The stock solutions were stirred for 24 hours (approx. 1110 rpm) with a magnetic stirrer. Thereafter, the stock solutions were used to prepare 5 test itejm concentrations (9.38 - 18.8 - 37.5 - 75.0 - 150 mg/L), which have been used for the algae toxicity test.

Results with reference substance (positive control):
The toxicity of potassium dichromate to Pseudokirchneriella subcapitata was determined over a period of 72 hours from 2014-10-14 to 2014-10-17. The reference item toxicity was in the valid range following SOPs from the test facility: ErC50 = 0.749 mg/L
Reported statistics and error estimates:
EC50-values of growth rate after 72 hours was calculated by sigmoidal dose-response regression.
NOEC/LOEC were determined by calculation of statistical significance of growth rate. As standard One Way Analysis of Variance (ANOVA) and DUNNETT's test were used for NOEC/ÖLOEC calculations. When running a One Way Analysis of Variance a Normality test and an Equal Variance test were done first. The SHAPIRO-WILK-Test was used to test for normally distributed populations. The Levene median test was used for equal variance. P-values for both Normality and Equal Variance tests are 0.05.

The toxicity of the test item to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to principles of OECD 201 and Council Regulation (EC) 440/2008/Method C.3.

The study was conducted under static conditions with an initial cell density of 7085 cells/mL. Five concentrations were tested in a geometrical series with a dilution factor of 2 (nominal): 9.38 - 18.8 - 37.5 - 75.0 - 150 mg/L. Three replicates were tested for each test item concentration and six replicates for the control. The environmental conditions were within acceptable limits.

The concentrations of the test item were analytically verified by LC-MS/MSat the start and at the end of the exposure of all test concentrations and the control. The measured concentrations of the test item at the start of exposure (0 hours) were in the range of 105 to 109% of the nominal values. The measured concentrations of the test item at the end of exposure (72 hours) were in the range of 95 to 106% of the nominal value. Therefore, all effect values are based on the nominal test item concentrations.
