Toxicokinetics

According to availble data [14C] DCY was rapidly distributed, metabolized, and excreted by rats. In 24 h after intravenous dosing, rats excreted 81.1% of the dose in the feces and 16.0% of the dose in the urine. For rats fitted with biliary cannulas, 54.5% of the dose, all of which was metabolites of [14C]DCY, was recovered in the bile 4 h. Associated with the rapid and extensive biliary excretion of metabolites of intravenously administered I14C]DCY was the appearance of large amounts of radioactivity in the feces and also, at intermediate time points, in the liver, gut contents, and gut tissue. In conclusion, rats rapidly distribute, metabolize, and excrete [14C]DCY. Since no toxic effects were evident, this study has provided no basis for assuming that exposure to small amounts of DCY would be hazardous to the health of humans.

Acute Toxicity (Oral, Dermal, Inhalation):

The available information indicates that the substance D&C Yellow No. 11 (CAS No 8003-22-3) is not likely to exhibit acute toxicity by the oral, inhalation & dermal route of exposure and hence is not classified as causing Acute Oral, Inhalation and Dermal toxicity according to CLP regulation.

Skin & Eye Irritation:

The available information suggests that the substance 1,3-isobenzofurandione, reaction products with methylquinoline and quinoline (D & C Yellow No. 11) is not classified as a skin and eye irritant according to CLP regulation.

Skin Sensitization:

The chemical D&C Yellow No. 11 is likely to be "sensitizing" to the skin of guinea pig and human as per the available experimental data. Thus, the chemical is classified as being "sensitizing”.

The available data (experimental and estimated) indicates that the test substance 1,3-isobenzofurandione, reaction products with methylquinoline and quinoline (Synonym: D & C Yellow No. 11) is not classified for acute toxicity via oral, inhalation & dermal route.

Available data also indicates that the chemical does is likely to exhibit genotoxicity. The chemical is not likely to be a reproductive toxin within the dose levels mentioned in the end points  

In the absence of local effects following short-term or long-term exposure, no dose-response data are available and a quantitative dose descriptor is not derived.
