PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution (100 mg/L) was prepared with dilution water one day prior to application and shaken with 20 rpm for 24 h at room temperature. After a separation phase of 1 h the WAF was taken from the homogeneous liquid phase.
- Eluate: Dilution water
- Differential loading: WAF with a nominal loading of 100 mg/L.
- Controls: Four replicates of dilution water without test item tested under the same conditions as the test groups.

Executive summary:

In the acute immobilization test with Daphnia magna (STRAUS) the effects of the water accommodated fraction (WAF)* with a nominal loading of 100 mg/L as a limit concentration of the test item Flue Dust S(batch number: Not specified) were determined according to OECD 202 (2004). The limit test was conducted under static conditions over a period of 48 h from 2010-03-15 to 2010-03-18 with the definitive exposure phase between 2010-03-16 to 2010-03-18 at Dr. U. Noack-Laboratorien, Käthe-Paulus-Str. 1, D-31157 Sarstedt, Germany.

20 test organisms were exposed to the water accommodated fraction, the pH-control and the control. No analytical determination of the test item concentration was carried out, because the test item is a mixture of various inorganic compounds with different properties.

The water quality parameters (pH-value and dissolved oxygen concentration), measured at 0 and 48 h, were determined to be within the acceptable limits. The validity criteria of the test guideline were fulfilled.

As no immobilization after 24 h and mean immobilization rates of The EC50-value for the reference test was calculated by using sigmoidal dose-response regression. The confidence interval for the EC50-value was calculated from the best-fit values, the standard error and the t-distribution with the software GraphPad prism.
The EL10-value after 48 h was calculated with the software GraphPad prism by using sigmoidal dose-response regression, respectively. The concentration effect relationship after 48 h is presented graphically.

Based on the nominal loading levels of the test item Flue Dust T the 48-h EL50 for Daphnia magna was estimated to be 100 mg/L.
The NOEL after 48 h was determined to be 50.0 mg/L as the highest tested loading level with no biologically significant effect (Immobilization rates of  10 % are seen to be not significant according to the guideline). The LOEL after 48 h was determined to be 100 mg/L as the lowest tested loading level with a biologically significant effect (Immobilization rates of > 10 % are seen to be significant according to the guideline).

In the acute immobilization test with Daphnia magna (STRAUS) the effects of the test item  Flue Dust T(batch number: 12-2009) were determined according to OECD 202 (2004) from 2010-03-15 to 2010-03-18, with the definitive exposure phase from 2010-03-16 to 2010-03-18, at Dr.U.Noack-Laboratorien, Käthe-Paulus-Str.1, D-31157 Sarstedt.

The study was conducted under static conditions over a period of 48 h with 5 water accommodated fractions (WAF)* with nominal loadings ranging from 6.25 to 100 mg/L set up in a geometric series with a dilution factor of 2. Additionally a pH-control was tested within the test series. 20 test organisms were exposed to each loading level, the pH-control and the control.

No analytical determination of the test item concentration was carried out, because the test item is a mixture of various inorganic compounds with different properties. All effect values, presented in Table 1, are based on the nominal loading levels of the test item Flue Dust T.

The water quality parameters (pH-value and dissolved oxygen concentration), measured at 0 and 48 h, were determined to be within the acceptable limits. The validity criteria of the test guideline were fulfilled.
