Description of key information
Reliable results are available from short-term tests with fish, aquatic invertebrates and algae. The LL50/EL50 values determined in these tests conducted with water-accommodated fractions (WAFs) of TOPP were 29 mg/l, 6.4 mg/l and 17.1 mg/l for Danio rerio (fish), Daphnia magna (invertebrate) and Desmodesmus subspicatus (algae) respectively. A No Observed Effect Loading Rate (NOELR) of 5 mg/l was also determined in the test with D. subspicatus. These results represent the lowest values obtained in two or three equally reliable tests carried out on different samples of the substance. Studies conducted using WAFs are aimed principally at the determination of the hazard properties of whole substance. For compositionally complex substances, such as TOPP, the concept of a single PNEC that reflects the toxicity of the whole substance is not appropriate because of variability in the properties of the constituents. Therefore, PNECs are obtained for the blocks of constituents (a block may comprise of one or more constituents) that make up the whole substance.
Read-across/supporting information is also available for the substance “ UVCBs hydrocarbons, terpene-processing by-products” (CAS 68956-56-9). The 96h-LL50 for Danio rerio was determined as 5.07 mg/l in a test conducted with WAFs.
Short-term toxicity data are also available for alpha- and beta-pinene, which are both constituents of block 1 of TOPP. For alpha-pinene the LC50/EC50 values were 0.30 mg/l for Danio rerio (fish) and 0.48 mg/l for Daphnia magna (invertebrate), and the EC50 value for beta-pinene with Daphnia magna was 1.25 mg/l. Both substances were also tested with algae (Pseudokirchneriella subcapitata). The 48h-LOEC for alpha-pinene was determined as 0.494 mg/l (and the NOEC was estimated to be around 0.25 mg/l) and the 48h-EC50 and 48h-EC10 for beta-pinene was determined to be 0.83 mg/l and 0.38 mg/l respectively. However, significant loss of test substance was observed in both tests (presumably a result of biodegradation in and/or volatilisation from the static test system) and so these values may not represent the true toxicity of alpha- and beta-pinene.
There are no measured data available for long-term toxicity of TOPP to fish or aquatic invertebrates. Data-waivers are considered to be appropriate for these endpoints on the grounds that calculation of PNECs for the aquatic compartment will be based on data for the blocks of constituents rather than on data for the whole substance. Testing for long-term toxicity to fish and invertebrates will therefore not contribute to the database that is required to complete the assessment of the substance.
