- Concentrations: The concentrations of CHPTAC and its conversion product 2,3-Epoxypropyltrimethylammonium chloride (EPTAC) were determined via HPLC/MS MS in all test item concentrations (0.17, 0.51, 1.52, 4.56, 13.7 mg/l) and controls.

- Sampling method: Samples were taken on days 0, 7, 14, 19 (fresh media) and days 2, 9, 16, 21 (old media).

PREPARATION AND APPLICATION OF TEST SOLUTION

- Method: a stock solution of 100 mg/L (i.e. 65.2 mg/L of the active ingredient CHPTAC) is said to be dispersed by agitation. No further details on the preparation of the test solution are reported. Given the high water solubility of the test substance it is assumed by the reviewer that either serial dilutions of the stock solution were made with test medium as dilution water, or that the test substance was added to the test medium to achieve the nominal concentrations desired.

- Source: the test organisms were bred at the testing facility, origin Insitut fur Wasser-, Boden- und Lufthygiene (WaBoLu).

- Age of parental stock (mean and range, SD): 2-24 h old, the test organisms were not the first brood progeny.

- Feeding during test

- Food type and amount: The food quantity was a D. subspicatus (0.395 - 1.45 mL) and C. vulgaris (0.375- 0.745 mL) suspension of algae per test vessel (corresponding to 0.1 - 0.2 mg C per Daphnia per day). lt varied according to the growth of the daphnids, but was the same for all test groups on any particular day.

- Frequency: At lest 5 times per week.


ACCLIMATION

- Acclimation conditions: the test organisms were bread in an incubator in similar conditions to the test (temperature, illumination and photoperiod), in 2-3 L glass vessels with approx. 1.8 L culture medium.

- Type and amount of food: a mix of D. subspicatus and C. vulgaris with algae cell density of >10^6 cells/ml ad libitum.

- Feeding frequency: at least 5x per week

- Health during acclimation: the stock population is defined as healthy by the authors of the report.

- Mortality of parent animals: 100% mortality occurred at 13.7 and 4.56 mg a.i./L treatments and 60% mortality at 1.52 mg a.i./L after 21 days.

- Body length and weight of parent animals: see Table 1 for details. No statistical analysis was applied to these parameters.

- Time to first brood release or time to hatch: no statistically significant difference between treatment levels and the control.

- Brood size: At 1.52 mg/l the reproduction output was significantly higher than in controls or other test groups indicating a stress reaction. No significant reduction in reproduction compared to the control was not observed at lower concentration levels.
