Undiluted 2-ethylhexanoylchloride (0.5 mL) was applied onto the skin of shaved White Vienna rabbits. Two animals were treated for 1, 5 and 15 minutes and 2 other animals for 20 hours using occlusive conditions. Erythema (24 h score 3/4) was observed following a 15-min occlusive exposure. Erythema (up to score 3) and edema (up to score 3) were observed after a 20 h exposure. For 20 h exposure, erythema (score 2) was still visible 8 days after treatment. The overall primary irritation score (intact skin) was 5.0/8.0 (20 h exposure) and 3/8.0 (15 min exposure), which was consistent with a rating of moderately irritating in both cases (BASF AG, 1968).

A Draize test was performed in which neat 2-ethylhexanoylchloride (0.5 ml) was applied onto two areas of shaved skin, one intact and one abraded, of 6 New Zealand White rabbits (Quinn & Paa, 1975). The occlusive dressings were removed after 24 hours. Skin was scored for erythema and edema (on a scale of 0-4) when the dressings were removed and again at 72 hours. Third degree chemical burns (on both intact and abraded sites) were found at 24 and 72 hours. The primary irritation score was 2.5/8.0 (24 h and 72 h observations combined; intact/abraded combined), which was consistent with a rating of moderately irritating.

Undiluted 2 -ethylhexanoylchloride (0.5 mL) was applied into the eyes of 2 White Vienna rabbits. The adjacent unterated eyes of the same animals served as control. The eyes were not washed out as recommended by the OECD TG 405. Eye reactions were read 24, 48 and 72 hours. The maximum score for opacity was 0.33 and for erythema 1.33. The effects were completely reversibele within 4 days (BASF AG, 1968).

2-Ethylhexanoylchloride (0.1 mL) was instilled into the conjunctival sac of the right eye of 6 New Zealand White rabbits. The procedure was according to Draize et al.1944. Eyes were not washed after instillation. Corneal opacity and iridal and conjunctival effects were evaluated 1, 24, 48 and 72 hours and 7 days after instillation of material. A maximum Draize irritation score of 13.0/110 was reached 1 h after application. Effects were fully reversible within 72 h (Quinn & Paa, 1975).

 

Justification for classification or non-classification

2-Ethylhexylchloroformates is irritating to the skin according to the classification criteria laid down in EU Regulation 1272/2008 (skin irr/corr Cat 2).

No classification is warranted concerning eye irritation as classification criteria EU Regulation 1272/2008 are not met.
