The registered substance is a complex petroleum UVCB whose constituents have variable physicochemical properties. Therefore, a modeling framework is needed to account for the bioavailability of the constituents in the mixture, which was then used to predict the toxicity of the substance as a whole.

The target lipid model (TLM) is a framework that relates toxicity to the physicochemical properties of a nonpolar organic constituent. The TLM, and HC5, have been applied to soils and sediment using equilibrium partitioning (EqP) model. This framework utilizes organic carbon partition coefficients (KOC) to convert aquatic based effect levels (CW) to bulk soil- and sediment-based effect levels. The combined TLM-EqP framework was validated previously for soil and sediment acute and chronic toxicity endpoints. The constituents in this substances are within the scope of the TLM and EqP frameworks. The model used for the calculations was the PETROTOX v4.0 model. PETROTOX is a well-documented and peer reviewed model that is widely used across the chemicals industry.
Principles of method if other than guideline:
The target lipid model (2015) and equilibrium partitioning model as described in the cited references were used to determine soil or sediment effect levels following an estimation of initial loading of the registered substance and its constituents in the substrate. PREPARATION OF SPIKED SEDIMENT
- Natural sediment from the same unpolluted site as the amphipods (organic content: 3.3%; silt fraction: 73%), homogenized before preparing the test sediment.
- Chemical name of vehicle (organic solvent, emulsifier or dispersant): acetone

- Spiked test sediment: 5 Ll acetone containing test substance added to 130 g pre-dried sediment. After the acetone had dried off in a fume cupboard, the sample was added to 460 g of damp sediment.

- Reference substance: fluoranthene dissolved in 5 mL of acetone added to app. 110 g pre-dried sediment.After the acetone had dried off in a fume cupboard, the sample was added to app. 510 g of damp sediment in a plastic bucket.

- The sediment in the blanks (with and without acetone) was treated like the test sediment but without adding test substance.

All sediments (test concentrations, reference substance and blanks) were prepared in plastic buckets. 130 mL of seawater was added to each bucket and the bucket were shaken (120 rpm) for 5 hrs.

Executive summary:

An acute toxicity test was conducted on the marine amphipod Corophium volutator. The test substance concentrations ranged from 126 to 1495 mg/kg sediment dw. The test sediment was from the same unpolluted site as the amphipods. The seawater was natural seawater from an unpolluted site at about 80 m depth in Byfjord, filtered through sand filter at the collection. The test substance was dissolved in acetone and added to the sediment as follows: 5 mL acetone containing the test substance was added to 130 g pre-dried sediment; after the acetone had dried off in a fume cupboard, the sample was added to 510 g of damp sediment in a plastic bucket, 130 mL seawater was added to the bucket and the bucket was shaken (120 rpm) for 5 hrs. The spiked sediment was then split into three beakers, 400 mL of seawater was added to each beaker, and they were aerated overnight before the test organisms were introduced. The blanks were prepared the same way (three with and three without acetone). In the test, 4 concentrations were tested in a geometric series: 126, 308, 668 and 1495 mg/kg. Three replicates were used for each concentration and each blank.

Dissolved oxygen was > 40% during the study. The pH was 8.1. The temperature was 15 +/- 2 °C.

The test organisms (5 -10 mm) were obtained from the same unpolluted site as the sediment.

No deviation from the protocol was noted. No analytical monitoring was performed.

The results mean that the 10-d LC50 is 503 mg/kg sediment dry weight.

Executive summary:

An acute toxicity test was conducted on the marine mollusc bivalve Abra alba. The test substance concentrations ranged from 5 to 118 mg test substance / kg 8% standard sediment suspension. Test medium were prepared using microencapsulation because of the hydrophobicity of the susbtance. The standard sediment used for microencapsulation (together with gelatine and acacia gum) and standard sediment suspension (8% (dw) standard sediment in natural seawater) was a natural sediment from a location where Abra alba were present, sieved on 45 µm mesh. Test systems consisted of polyethylene tubes (inner diameter: 36 mm; outer diameter: 40 mm) with 90 µm mesh, glued together in 3x5 racks and introduced in containers containing 250 g of standard sediment suspension. 5 concentrations were tested: 5, 10, 20, 52, and 118 mg test substance / kg 8% (dw) standard sediment suspension. Two replicates for the test substance and four replicates for the control were used, with 1 organism per tube (15 organisms per container).

The temperature was 11 +/- 1 °C. Analytical monitoring was not performed.

The test organisms were acclimated for 5 days prior to the exposure. A 2-d pre-exposure was performed to establish the normal fecal pellet production before exposure.

An acute toxicity test was conducted on the marine mollusc bivalve Abra alba. The test substance concentrations ranged from 27 to 611 mg test substance / kg 8% standard sediment suspension. Test medium were prepared using microencapsulation because of the hydrophobicity of the susbtance. The standard sediment used for microencapsulation (together with gelatine and acacia gum) and standard sediment suspension (8% (dw) standard sediment in natural seawater) was a natural sediment from a location where Abra alba were present, sieved on 45µm mesh. Test systems consisted of polyethylene tubes (inner diameter: 36 mm; outer diameter: 40 mm) with 90 µm mesh, glued together in 3x5 racks and introduced in containers containing 250 g of standard sediment suspension. 5 concentrations were tested: 27, 59, 119, 240, and 611 mg test substance / kg 8% (dw) standard sediment suspension. Two replicates for the test substance and four replicates for the control were used, with 1 organism per tube (15 organisms per container).

Standard sediment suspension with 8% sediment particle content (dry weight) and particle size <= 45µm ,and with a salinity of 31-35 per thousand, contaminated with test substance, stored 4-16 hrs at 11 °C under continuous aeration before exposure.
The test substance was a liquid insoluble in water. A 1/100 stock solution was prepared using microencapsulation. The test substance was microencapsulated to stock solution together with sediment particles. The microcapsule wall (gelatine + acacia gum) is permeable to small molecules and water soluble components will enter the water phase until equilibrium; The final test media are prepared in 500 ml glass bottles, which are shaken vigourously by hand for 30 seconds before transfer to the test aquaria.
