PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The test item was not soluble in tap water at the required concentration and no stock solution could be prepared. Therefore, 501.9 mg of the test item was weighed by means of an analytical balance and transferred to the test flask with 284 mL of tap water. The test item was mixed into the tap water by ultrasonic treatment for ten minutes and intense stirring for 24 hours at room temperature in the dark to dissolve a maximum amount of the test item and/or disperse it as homogeneously as possible. No emulsifiers or solvents were used. After the stirring period, 16 ml of synthetic wastewater and 200 ml of the activated sludge inoculum were added.

EFFECT PARAMETERS MEASURED : For measurement of the respiration rate, a well-mixed sample of each test medium was poured into a BOD-flask after exactly three hours incubation time, and was not further aerated. Then, the dissolved oxygen concentration was measured with an oxygen electrode (WTW TriOxmatic 300 and an oxygen meter WTW Oxi 539, Wissenschaftlich-Technische Werkstaetten WTW, Weilheim/Germany), and was continuously recorded for about ten to fifteen minutes. During measurement, the samples were continuously stirred on a magnetic stirrer. The oxygen consumption rate (in mg O2 L-1 minute-1) was determined from the linear part of the respiration curve in the range of 6.5-2.5 mg O2/L. I n case of very rapid oxygen consumption, the range used was below the limits indicated above, but always within the linear part of the respiration curve. In case of low oxygen consumption, the rate was determined over a period of at least ten minutes.

Executive summary:

The inhibitory effect of cerium dioxide on the respiration rate of aerobic wastewater microorganisms of activated sludge was investigated in a 3-hour respiration inhibition test according to the EU Commission Directive 88/302/EEC, Part C.11, and the OECD Guideline for Testing of Chemicals, No. 209.

A limit test was performed with one concentration of cerium dioxide of nominally 1003.8 mg/L.

In addition, two controls and three different concentrations of the reference item 3,5 dichlorophenol (5, 16, and 50 mg/L) were tested in parallel. After a 3-hour incubation time, the inhibition of the respiration rates was investigated via a determination of oxygen consumption rates.

The results of the reference treatments (EC50 = 12 mg/L within the guideline-recommended range of 5 - 30 mg/L) confirmed suitability of the activated sludge and the method used.

The test item cerium dioxide had no significant inhibitory effect on the respiration rate of activated sludge after the incubation period of three hours at the limit test concentration of 1003.8 mg/L. Thus, the 3-hour NOEC of cerium dioxide to activated sludge microorganisms was > = 1003.8 mg/L. The 3-hour EC20, EC50 and EC80 were clearly higher than 1003.8 mg/L.

Description of key information

By analogy with one of its constituents (i.e. cerium dioxide), the reaction mass of cerium dioxide and zirconium dioxide is not expected to give rise to adverse effects in aquatic microorganisms. Therefore, no study on the reaction mass is considered necessary.

Additional information

No data is available on the reaction mass. Based on the physicochemical, environmental and ecotoxicological data available on both the reaction mass and its two constituents cerium dioxide and zirconium dioxide, the three substances show highly similar intrinsic properties. Therefore, data available for the constituent cerium dioxide are included in the dossier as supporting information for the waiving of further studies with the reaction mass.

Micrometric cerium dioxide: Two studies are included. One of them (Grutzner, 2006) is quoted as reliability 1 according to Klimisch and the second was given a reliability 2 (Park et al, 2007). Both studies concluded that cerium dioxide was not harmful to aquatic microoorganisms, as no adverse effects were observed at the highest dose tested (1000 mg/L).

Nanometric cerium dioxide: One literature source, scored as Klimisch 2, is available concerning the nanoparticulate form (Park et al., 2007). It concluded that nanometric cerium dioxide was not harmful to aquatic microorganisms, as no adverse effects were observed at the highest dose tested (1000 mg/L).
