Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Description of key information

The key value of toxicity of 1-chlorobutane on microorganisms was determined as EC50(3h) of >1000 mg/L.

Key value for chemical safety assessment

EC50 for microorganisms:
1 000 mg/L

Additional information

The key study for acute toxicity of 1-chlorobutane to activated sludge was conducted according to OECD-guideline 209 and to GLP under static conditions (Evonik-Degussa 2010). The effect of a concentration range of 10 to 1000 mg/L of test substance on the respiration rate of 9.6 mL synthetic sewage in a 120 mL inoculum suspension after 3 hours resulted in an EC50(3h) of >1000 mg/L.

Three supporting publications evaluated effects on Photobacterium, Nitrosomonas, methanogens, aerobic heterotrophs, Vibrio and Shk1 with resulting I/EC50 values ranging from 110 to 486 mg/L.

(Blum et al. 1991: Photobacterium IC50(5min)=480 mg/L; Nitrosomonas IC50(24h)=120 mg/L; methanogens IC50(96h)=110 mg/L; Aerobic heterotrophs (Microtox) IC50(49h)=230 mg/L; Ren et al. 2003, Shk1 EC50=278 mg/L; Ren & Frymier 2002, Shk1 calculated from log(1/EC50) in mM: EC50calc.(5 min) = 279.6 mg/L; Vibrio fisheri (Microtox assay) EC50calc.(5 min) = 485.8 mg/L)
