Materials and methods
Test guideline
- Qualifier:
- equivalent or similar to guideline
- Guideline:
- OECD Guideline 404 (Acute Dermal Irritation / Corrosion)
- Deviations:
- yes
- Remarks:
- - Animals were treated for 1, 5, and 15 min or 20 hours using occlusive conditions; - Skin tissue from the ear was also tested; - The test substance was washed out with water (which sometimes contained a mild detergent) after the exposure period.
- Principles of method if other than guideline:
- BASF-Test
- GLP compliance:
- no
Test material
- Reference substance name:
- Dimethyl sulphate
- EC Number:

- Cas Number:
- 77-78-1
- Molecular formula:
- C2H6O4S
- IUPAC Name:
- dimethyl sulfate
- Details on test material:
- - Name of test material (as cited in study report): dimethyl sulfate
- Physical state: liquid
Constituent 1
Test animals
- Species:
- rabbit
- Strain:
- Vienna White
- Details on test animals or test system and environmental conditions:
- TEST ANIMALS
- Age at study initiation: young adult animals
- Weight at study initiation: 2.35 kg (female; 2.46 kg at the end of the study) and 2.5 kg (male; 2.36 kg at the end of the study)
Test system
- Type of coverage:

- Preparation of test site:
- other: clipped and intact skin
- Vehicle:
- unchanged (no vehicle)
- Controls:
- other: untreated skin of the same animal
- Amount / concentration applied:
- TEST MATERIAL
- Amount(s) applied (volume or weight with unit): 0.5-1 ml - Duration of treatment / exposure:
- 1 min, 5 min, 15 min and 20 hours
- Observation period:
- 8 days
- Number of animals:

SCORING SYSTEM:
Dermal reactions were classified using a BASF-scoring system, which is convertible into the current OECD Draize scheme.
Results and discussion
In vivo
Results
- Irritation parameter:
- other: the test substance is corrosive
- Basis:
- mean
- Reversibility:
- not reversible
- Remarks on result:
- other: necrosis was still observable at the end of the exposure period (8-day)
- Irritant / corrosive response data:
- Short time exposure caused no effects (1 min exposure) or light erythema (score: 2; 5 and 15 min exposure): Cutaneous reactions were reversible after 1 and 5 min exposure, but not after 15 min exposure at the end of the observation period.
Long time exposure (20 hours): Severe cutaneous reaction (erythema score 4 and edema score 3) and severe necrosis of skin tissue was reported for both animals 24 hours after application of the test substance, which persisted until the end of the observation period (8 days after application of the test substance).
Any other information on results incl. tables
Conclusion:
The test substance is considered to be corrosive based on the irreversible necrosis
