The long-term toxicity of Guanidine mononitrate (CAS 506-93-4) to aquatic invertebrates was investigated in a study (van der Schalie 1985) using reproduction of daphnids (Daphnia magna) comparable to the OPPTS 850.1300 (1996) and OECD TG 211 (1998) protocols. GLP is not stated. The experiment is deemed valid, conclusive and thus suitable for assessment with restrictions.

The test animals were exposed during 21 days under flow through conditions to control and test item concentrations of nominal 60, 33.6, 18.8, 10.5, and 5.9 mg/L (measured 52, 26.2, 16.8, 8.8, 4.2 mg/L) in the first test and 6.19, 3.45, 1.97, 1.08, 0.63 mg/L (measured 6.09, 2.90, 1.66, 1.14, 0.75 mg/L) in the second test respectively. In the first test significant toxic effects were found at all treatments. Therefore a second, definitive test was performed with lower concentrations. The effects were related to the measured concentrations. In test system (Test 2) was maintained at 19.4-20.6 °C, 7.6 to 8.7 mg/L mg dissolved oxygen/L and a pH of 7.8 to 8.2. The hardness of the test media were 165 to181 mg/L CaCO3, which corresponds to more than ca. 99-109 mg carbonate/L. The effect parameters measured were survival, total young per replicate tank, growth reproduction rate, immobilization and the number young per female per replicate day, which was the most sensitive endpoint, different from control at the highest tested level.

Accordingly the 21-d NOEC and LOEC-values, based on reproduction were 2.9 and 6.09 mg test item/L respectively. The NOEC corresponds to 23.7 µmol Guanidinium kation/L or considering equimolar conversion to this species 2.14 mg Guanidine carbonate (CAS 593-85-1)/L.

In conclusion the test item (source chemical) was found not being toxic to aquatic invertebrates up to the relevant levels considered for long-term hazard to the aquatic life in the EU (i.e. 1 mg/L according to CLP, Regulation (EC) No 1272/2008)) and no aquatic hazard classification is required the results of this study. This conclusion can be read-across to the target chemical Guanidine carbonate.
