In accordance with column 2 of REACH Annex IX, the bioaccumulation testing for COASOL (required in section 9.3.2) is not proposed as the substance has a low potential for bioaccumulation.COASOL is comprised of diisobutyl esters (di-isobutyl succinate, di-isobutyl glutarate and di-isobutyl adipate), which will hydrolyze when they are taken up by an organism. This releases isobutanol and then adipic, glutaric and succinic acid, which are all endogenous in mammals. Succinic acid is a part of the citric acid cycle and so will be taken into the organisms normal metabolic processes, glutarate can be further converted into alphaketo-glutarate and enter the citric acid cycle, and adipate can be converted into a number of things, such as urea, or excreted as is. In addition, results from QSAR modelling (BCFBAF) of the individual constituents (di-isobutyl succinate, di-isobutyl glutarate and di-isobutyl adipate) using the Arnot-Gobas model, which incorporates metabolism and bioavailability of the substances, estimates BCF values for all constituents of <13 L/kg wwt. Hence, the potential for bioaccumulation is minimal and testing will be waived.
