- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution with a nominal concentration of 10 mg/L was prepared by adding the necessary amount of test item to a volumetric flask. Test medium was added and the solution was stirrered for 60 minutes on a magnetic stirrer at 750 rpm. The solution was slightly turbid and foam was observed. The highest test concentrations was prepared under stirring. Lower test item concentrations were prepared by dilution with test medium. Additional information
The chronic toxicity of Alcohols, C12-15, branched and linear, ethoxylated to aquatic invertebrates was investigated in a GLP guideline study following OECD GD 211. The effect of the test substance on the reproductive output of Daphnia magna was assessed in a semi-static test with a daily renewal of test solutions. Daphnids were exposed to test substance concentrations of 0.320, 0.100, 0.032, 0.0100, 0.0032 and 0.0010 mg/L. The test concentrations were analytically monitored by HPLC-MS/MS by analysing the concentration of two representative constituents. The time weighted mean concentrations of the four highest test concentrations were 0.0792, 0.0356, 0.0956, and 0.00431 mg/L. The two lowest test concentrations could not be detected analytically. No statistically significant mortality was determined at any of the test item concentrations. Based on the number of living offspring produced per surviving parental animal a NOEC (21 d) of 0.0356 mg/L (TWA) was determined.
