Key value for chemical safety assessment

Additional information

The acute toxicity of dimethylammonium 2,4-dichlorophenoxyacetate (CAS No. 2008-39-1) was investigated by Szentes (2001). This test was performed according to US EPA 850.2100 Avian Acute Oral Toxicity test (1996) and GLP. A single dose of the test item was administered by oral gavage to 50 birds (10 birds per dose group), followed by an observation period of 14 days. Mortality was reported only at the highest concentration tested (900 mg test substance/bw.kg), whereas clinical symptoms (ruffled feathers, reduced activity, etc) were observed at the 323, 539 and 900 mg/bw.kg up to 3 days post-treatment. From that moment onwards, those symptoms remitted completely. Feeding consumption and body weight were not affected by treatment with the test item, except from values between days 0-3 at the 900 mg/bw.kg dose. The resulting LD50 was > 900 mg test substance/bw.kg (corresponding to 367.5 mg a.i./bw.kg) and the NOEL based on body weight and clinical observations, 193 mg test substance/bw.kg (78.8 mg a.i./bw.kg). Additional information on the acute toxicity of dimethylammonium 2,4-dichlorophenoxyacetate to birds could be gathered (Hoxter et al., 1990). This test, conducted according to EPA/FIFRA Guideline 71-1 on Colinus virginianus, resulted in a LD50 of 500 mg a.i./kg bw/day.

Additionally, the dietary toxicity of dimethylammonium 2,4-dichlorophenoxyacetate was determined in a study by Gaborné (2001), conducted according to OECD Guideline No. 205: Avian Dietary Toxicity Test (1984), under GLP conditions. The test organisms (10 per replicate,Coturnix coturnix japonica) were fed for 5 days poultry standard diet, treated with the test item. Mortality, clinical symptoms, feed consumption and body weight were measured throughout the exposure and post-observation periods (3 days). No effects were reported, except for a reduction in body weight at the highest concentration tested (4028 mg/kg food) compared to control group values. Therefore, the LC50 value was determined to be over 4028 mg test substance/kg food (1644 mg a.i./L)
