Description of key information

As no data on toxicokinetic are available for the registered substance, for chemical safety assessment purposes, the following worst-case assumptions are made:


The molecular weight of CARDOLITE NC-513 is 356.54 g/mol and thus indicates a potential for oral absorption. However, due to its high log P value of (> 6.2, OECD 117) and its only low water solubility (0.89 – 1.77 mg/L at 20.0 ± 0.5 °C, OECD 105), absorption may be limited. Nevertheless, for chemical safety assessment purposes an oral absorption rate of 100 % is assumed following a worst-case approach.


The ECHA guidance criteria (Chapter R.7C) state that 10% dermal absorption is used when the molecular weight of the substance is >500 and the log Pow is <-1 or >4, otherwise 100% dermal absorption is used. Thus, based on the physical-chemical properties indicated above, the dermal absorption of the registered substance is considered to be 100 %.


An inhalation absorption rate of 100% is considered, as there is no data on absorption via inhalation available. Furthermore, it should be noted that for chemical safety assessment purposes, the absorption rate of the starting route is considered to be by a factor of 2 lower than the one of the end-route in case of route-to-route extrapolation from oral to inhalation following a worst-case approach (based on ECHA Guidance R.8).
