- Sampling method: Analytical samples taken at respective time points during the study course (exposure start, day 1 (aged), day 7 (fresh), day 8(aged), day 16 (fresh), day 17 (aged), day 20 (fresh), day 21 (aged))
- Sample storage conditions before analysis: The samples were mixed with 3 mL toluene and shaken by hand for 60 seconds. In the analytical laboratory, the toluene phase was separated (for aged samples sodium chloride was added for better phase separation, partly the extracts were centrifuged at 1800 rpm for 2 min.) and stored deep-frozen (≤ - 18 °C) until analysis.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution with a nominal concentration of 25 mg/L was prepared by adding the necessary amount of test item to a volumetric flask. Test medium was added and the solution was homogenised by shaking. The solution was observed to be clear and transparent and some foam was observed. Test item concentrations were prepared by dilution with test medium.

- Method of breeding: Daphnia magna was bred as single culture (1 daphnid per 100 mL) in Elendt M4 medium. pH-value: 6.0 – 9.0, dissolved oxygen: > 60 % saturation, total hardness 140 - 250 mg/L (as CaCO3), fed with single cell green algae (Desmodesmus subspicatus, formerly Scenedesmus subspicatus) at least three times a week.

- Other biological observations: At nominal test item concentrations 2.38 to 78.1 µg/L no mortality above the allowed control mortality was observed. At the highest nominal test item concentration of 250 µg/L 30 % of the adult daphnids were dead at the end of exposure

The NOEC and LOEC were determined by using a multiple comparison method (Fisher’s exact test for mortality of adult daphnia, multiple Welch’s t-test with Bonferroni-Holm adjustment for alive offspring per adult from test start, Williams’ test for alive offspring per alive adult at test end). The EC20, 50- value for alive offspring per alive adult from test start and alive offspring per alive adult at test end was determined with 3 parameter logistic cumulative distribution function. The statistical program was not able to derive EC10-value of reproduction based on the data presented. Due to a weak dose response for EC20-values no reliable values were calculable and the EC20-values were not applicable. The LC50 for mortality of adult daphnia was not determined statistically.

Additional information

The chronic toxicity Alcohols, C12-13, branched and linear, ethoxylated to Daphnia magna was investigated in a GLP guideline study following OECD 211. The study was conducted under semi-static conditions with a daily renewal of test solutions. During the test period of 21 days the mortality of test organisms, time of first production of offspring and the number of offspring were assessed. The test organisms were exposed to nominal test item concentrations of 0.00238, 0.00763, 0.0244, 0.0781, and 0.250 mg/L. The test concentrations were analytically monitored by HPLC-MS/MS. The time weighted mean measured concentrations, based on three representative constituents of the UVCB substance, were 0.0019, 0.00437, 0.0123, 0.0401 and 0.131 mg/L. Based on the number of living offspring produced per surviving parental animal a NOEC (21 d) of 0.0123 mg/L (TWA) was observed.
