Description of key information

1,2-Methylen-4,6-dienacetate (ZK 5690) is an intermediate of the synthesis of cyproteronacetate (CAS 427-51-0) and structurally very closely related. Due to its predominantly apolar components the substance is lipophilic in accordance with a log Pow of 2.66.

Cellular membranes are therefore expected to be penetrated and a dermal as well as an oral resorption is anticipated. In analogy to cyproteronacetate a substantial resorption from the GI tract is expected (see public informations on Androcur ® , 1991).

The substance is expected to be completely excreted in form of metabolites by both the biliary and the renal pathway.

Metabolism in analogy to other endogenous steroid hormones will predominantly take place in the liver such as reduction of the A -ring and the keto-group, and hydroxylation such as 15-beta-hydroxylation of cyproteronacetate.
