Test material

Constituent 1
Chemical structure
Reference substance name:
Aluminium, compound with nickel (1:1)
EC Number:
234-439-6
EC Name:
Aluminium, compound with nickel (1:1)
Cas Number:
12003-78-0
Molecular formula:
Al.Ni
IUPAC Name:
aluminum - nickel (1:1)
Test material form:
solid: particulate/powder
Details on test material:
dark grey fine powder

Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
For each treatment, 400 µL of the respective test item solution was mixed with the necessary amaount of algal pre-culture (1.85mL) to achieve a cell concentration of 4.6 x 10^3 cells/mL. In this mixture, the pH value was measured and samples for the analytical determination were taken.
The concentration of Al and Ni in the test vessels as contents of the test item were measured at the start and the end of the test.

Test solutions

Vehicle:
yes
Remarks:
demineralised water
Details on test solutions:
The nominal load of the highest concentration (32mg/L) was mixed with the corresponding amount of demineralised water and stirring vigorously for 96 +/- 3h. The resulting solution was filtrated through 0.45 µm nylon filters. The lower concentrations were prepared by dilution with demineralised water.

Test organisms

Test organisms (species):
Desmodesmus subspicatus (previous name: Scenedesmus subspicatus)
Details on test organisms:
Unicellular green algae, Desmodesmus subspicatus Strain 86.81, obtained in January 2016 from MEM Sciencebridge GmbH (Institut für Pflanzenphysiologie, Universität Göttingen) and kept as stock culture.

Study design

Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
72 h
Post exposure observation period:
no

Test conditions

Test temperature:
22.0 - 22.2°C
pH:
7.6-7.9
Nominal and measured concentrations:
1rst, 2nd, 3rd experiment: (nominal concentrations): 32/10/3.2/1/0.32 mg/L
4th, 5th experiment: (nominal concentrations): 10/3.2/1/0.32/0.1/0.032/0.01 mg/L
The determination of Al and Ni did not allow the calculation of the test item concentration, but showed clearly the presence of the test item in the solutions.
Details on test conditions:
5 experiments were performed.
The test vessels (glass flasks 65 ML total volume) were filled with 45 mL of the respective test solution and incubated open (covered with perforated plastic foil acting as a stopper) at light (4500 Lux) for 72h, shaken on an orbital shaker to keep algae in suspension. Before the start and every 24h, the cell number was determined by an electronic particle counter. After the end of the test (72h) the algae were examined microscopically.
Reference substance (positive control):
yes
Remarks:
Potassium dichromate, negative control: blank - nutrient medium

Details on results:
As a worst case scenario the results of the last (5th) experiment due to the strong inhibition of algal growth were taken for evaluation of the biological results. As inhibition of algal growth was only observable in the two highest treatments, only the three highest treatments were used for evaluation of the biological results.
Results with reference substance (positive control):
The 72h EC50s of potassium dichromate were determined in a seperate reference test. The values lay within the range of the laboratory: Growth rate: 0.73-1.10 mg/L, Yield: 0.21 - 0.66 mg/L
