In different study,2-methyl-4-nitroaniline (99-52-5)has been investigated for potential for dermal sensitization to a greater or lesser extent. The prediction is based on in vivo experiments in guinea for target chemical2-methyl-4-nitroaniline (99-52-5)and its structurally similar read across substances 2-Methoxy-4-nitroaniline(99-52-9). The predicted data using the OECD QSAR toolbox have also been compared with the experimental data of read across.

The skin sensitization potential of 2-methyl-4-nitroaniline (99-52-5) was estimated by SSS (2017) using OECD QSAR toolbox v3.3 with log kow as the primary descriptor and considering the six closest read across substances 2-methyl-4-nitroaniline (99-52-5) was predicted to be not sensitizing to the skin of Pirbright-White guinea pig.

Supported by experimental data conducted by U.S. (NTP) (Prepared for NCI to support chemical nomination by Technical Resources International, 2006) on structurally similar read across substance2-Methoxy-4-nitroaniline (99-52-9)in Guinea pig. The read across substances share high similarity in structure and log kow .Therefore, it is acceptable to derive information on skin sensitization from the analogue substance.The skin sensitization study of 2-Methoxy-4-nitroaniline (99-52-9) was performed in Dunkin Hartley guinea pig by Guinea pig maximization test .In induction phase, for intradermal injection7.5 percent (w/w) solution inAlembicol D, a coconut oil triglyceride and for dermal application 75% in Water concentration were used.In challenge phase 75% and 40% solution in distilled water was used .No evidence of sensitivity was noted. Hence it is considered that 2-Methoxy-4-nitroaniline (99-52-9) was not skin sensitizing in guinea pig.

 Thus based on the above predictions on2-methyl-4-nitroaniline (99-52-5) aswell as its read across substances and applying weight of evidence, it can be concluded that2-methyl-4-nitroaniline (99-52-5) isnot a skin sensitizer. Thus comparing the above annotations with the criteria of CLP regulation,2-methyl-4-nitroaniline (99-52-5)) can be considered as ‘not classified’ for skin sensitization effects.
