- Other:During the first definitive test, measurements of organic carbon showed significant differences between the two preparations. In order to improve preparation and to achieve a better homogeneity of the test solutions, the preparation of the stock solutions of the test item (1000 mg/L) was slightly modified. The ultrasonic treatment was conducted in two stages instead of one as indicated in the study plan. This modification allowed to reduce the disparity between preparations (Test N°1: 176.9 and 316.9 mg/L DOC and Test N°2: 293.3 and 217.7 mg/L DOC).

Sensitivity of the test organism was checked using potassium dichromate.
The test carried out from 12 to 13 March 2009 gave the following results:
EC50 24h: 1.27 mg/L (1.18 – 1.36 mg/L).
The EC50 24 hours was calculated using a logistic regression analysis with a bootstrap estimation of the 95% confidence limits.
The EC50 24h is in the recommended range: 0.6 – 2.1 mg/L. This result confirms the appropriate sensitivity of the strain of Daphnia magna.
TEST ITEM PREPARATION
The hydrosolubility of diester d’isosorbide is equal or lower than 5 mg/L.
In order to test the dissolved part of the test item, a stock solution at 1000 mg/L was prepared, for each renewal, in dilution water as follows:
1/ Ultrasonication of the test item (1 g) during 10 minutes in about 500 mL of dilution water
2/ 450 mL are collected in a 1 liter flask
3/ Ultrasonication of the 50 mL residual solution during 2 minutes in about 100 mL of dilution water
4/ 150 mL are collected in the 1 liter flask and completed up to 1 liter with dilution water
5/ Magnetic stirring during 10 minutes
6/ Centrifugation for 30 minutes (3000 g)
The stock solution was then diluted in dilution water to get diluted solutions at 100 and 10 mg/L to achieve the range of concentrations necessary to perform the test.
The test solution was renewed after a period of 24 hours.
