PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: For test concentrations of 10, 32, 100, 320 and 1000 mg/L, test item was dispersed in approximately 250 mL of water and subjected to ultrasonication for approximately 15 minutes followed by prolonged mixing for 24 h in order to maximise the dissolved test item concentration. For test concentrations of 1.0 and 3.2 mg/L, test item was dissolved in acetone followed by dispersion in test medium.
- Chemical name of vehicle (organic solvent, emulsifier or dispersant): Acetone for test concentrations 1.0 and 3.2 mg/L (to avoid very small amounts of test item being weighed out); acetone was also added to other test concentrations vessels in order to maintain consistency.
- Evidence of undissolved material (e.g. precipitate, surface film, etc): After 30 min. and 3 h contact time, the test concentrations of 1.0, 3.2, 10 and 32 mg/L contained a dark brown dispersion with no undissolved test item visible and the test concentrations of 100, 320 and 1000 mg/L contained a dark brown dispersion with globules of test item on the surface and dispersed throughout.

EFFECT PARAMETERS MEASURED (with observation intervals if applicable): The rate of respiration measured using a Yellow Springs dissolved oxygen meter fitted with a BOD probe. The rate of respiration was measured over the linear portion of the oxygen consumption trace (where possible
between approximately 6.5 and 2.5 mg O2/L). In the case of low oxygen consumption, the rate was determined over an approximate 10 min. period.

Results with reference substance (positive control):
The EC50 value for 3,5-dichlorophenol (8.4 mg/L; C.I.: 6.6-11 mg/L) lie within the acceptable range (5 to 30 mg/L) thus fulfilling the validity criteria.
Reported statistics and error estimates:
95% confidence limits were calculated for the reference item EC50 value using the method of Litchfield and Wilcoxon.

Control results: Variation in respiration rates of two controls was ± 8% (i.e. < 15%) thus fulfilling the validity criteria.

Other: In some instances, the initial and final dissolved oxygen concentrations were below recommended values (6.5 and 2.5 mg O2/L, respectively). This was considered to have had no adverse effect on the results of the study given that in all cases the oxygen consumption rate was determined over the linear portion of the oxygen consumption trace.
