Test solutions

Vehicle:
yes
Details on test solutions:
The dilution water used was soft water reconstituted fromdeionized water. The test solutions were prepared by the direct addition of the appropriate quantity of test substance and a proportional ammount of acetone to 15 liters of dilution water.

Test organisms

Test organisms (species):
Lepomis macrochirus
Details on test organisms:
The bluegill (Bionomics lot # 85A33b) were obtained from a commercial supplier in Missouri. The mean wet weight of the test fish population was 0.52 g (range 0.30 - 0.67 g, N=30) and the mean total length was 36 mm (range 27-40 mm, N=30). The fish were maintained in the lab at least 14 days prior to the test. They were fed a dry commercial pelleted food, ad libitum, daily except during the 48 hours prior to testing. There was no mortality of the test fish population during this 48-hour period.

Test conditions

Hardness:
41 mg/L as CaCO3
Test temperature:
22 + or - 1C
pH:
7.2
Nominal and measured concentrations:
700, 420, 250, 150 and 91 mg/L nominal
Details on test conditions:
The toxicity test was conducted in 18.9-L glass aquaria which contained 15L of test solution. The test solution depth was 18 cmwith a surface area of 985 cm2. Two control aquaria were established containing the same dilution water and maintained under the same conditions as the test aquaria containing the test substance. One control aquaria contained the greatest amount of acetone vehicle present in any test vessel (0.33 mg/L) and was designated the solvent control. The test temperature was maintained at 22 + or - 1C and the solutions were not aerated. The photoperiod was controled as 16 hours light and 8 hours darkness. Ten bluegill selected impartially from the holding tank were placed in each test aquaria within 20 minutes after the test solutions had been prepared. The resulting test organism loading concentration was 0.35 g of biomass per liter of test solution. The fish were not fed during the exposure. All aquaria were examined after 0, 24, 48, 72 96 hours of exposure as follows: mortalities were recorded, dead fish were removed, and observations of the fish and physical characteristics of the test solutions were recorded.

Details on results:
No mortality was observed in the control, solvent control, and test substance concentrations up to and including 420 mg/L. In the 700 mg/L test concentration 20% mortality was observed at 24 hours and ddi not increase over the remaining exposure period.

Applicant's summary and conclusion

Executive summary:

A 96 -hour static acute toxicity tests was conducted with TMPD using bluegill (Lepomis macrochirus) utilizing a method equivilent to U.S. Environmental Protection Agency Methods for Acute Toxicity Tests with Fish, Macroinvertebrates & Amphibians. The exposure was conducted using nominal concentrations of the test substance at 91, 150, 250, 420 and 700 mg/L. The LC50 was determined to be > 700 mg/L, the highest concentration tested. The NOEC was 420 mg/L.
