Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Key value for chemical safety assessment

Genetic toxicity in vitro

Description of key information

In a reliable and GLP guideline study, the substance showed no evidence of mutagenicity in a bacterial reverse mutation (Ames) assay in Salmonella typhimurium strains TA98, TA100, TA1535 and TA1537 and Escherichia coli strain WP2uvra, with or without metabolic activation. An older study using bacterial strains TA98, TA100, TA1535 and TA1537 and 87% pure 2-(2-(2-butoxyethoxy)ethoxy)ethanol similarly did not cause an increase in mutations when tested at up to 5mg/plate with and without S9 metabolic activation using both a standard plate incorporation and pre-incubation methods of application.

In a guideline and GLP in vitro chromosome abberation study using human blood lymphocytes, 2 -(2 -(2-butoxyethoxy)ethoxy)ethanol did not induce chromosomal aberrations in human lymphocytes with our without metabolic activation.

In a guideline and GLP HPRT mammalian cell gene mutation assay using Chinese Hamster lung fibroblasts, the substance 2 -(2 -(2 -butoxyethoxy)ethoxy)ethanol was unable to induce gene mutations and therefore considered negative in the assay.

Endpoint conclusion

Justification for classification or non-classification

From the available data, there is no indication of the substance having genotoxic or mutagenic properties, therefore classification is not warranted.
