Additional information

Since no data on the toxicity of 2-methoxyethyl acrylate (CAS 3121-61-7) to aquatic algae and cyanobacteria are available, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 read-across to another structurally related analogue substance was conducted. Read-across data from ethyl acrylate (CAS 140 -88 -5) were used, which was considered suitable for read-across.

The key study was conducted with ethyl acrylate according to GLP and OECD guideline 201 using Selenastrum capricornutum as test organism (ABC Laboratories, 1990). Within this study, nominal test concentrations of 6.5, 13, 25, 50 and 100 mg/L were applied with corresponding mean measured concentrations of 5.9, 11, 23, 47 and 99 mg/L at test start (recovery 92 ± 5.1 % of nominal concentration). After 96 h, the measured concentrations had decreased below the detection limit (three lowest concentrations) or below 1% of the nominal concentrations (two highest concentrations). It is discussed that the decreased concentrations may be due to 1) volatility of the compound, 2) adsorption to the glass aquaria, and/or 3) adsorption to particulate, including algal cells, in the test solutions. Thus, a recalculation using the arithmetic means for the exposure concentrations was conducted. As a result, a 72h-EC50 value of 4.5 mg/L for cell number was determined based on measured concentrations (arithmetic mean) (ABC Laboratories, 1990). Cell number/growth was significantly inhibited at all concentrations from 24 through 96 hours, thus the determination of a NOEC value was not feasible.

Due to structural and property similarities with the tested analogue substance, it can be concluded that the above result also applies to 2-methoxyethyl acrylate (CAS 3121-61-7). For details see analogue justification in the overall summary of Chap 6.1 and in Chapter 13 of the Technical Dossier.
