Since no data on the short-term toxicity of 2-methoxyethyl acrylate (CAS 3121-61-7) to fish are available, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 read-across to another structurally related analogue substance was conducted. Read-across data from ethyl acrylate (CAS 140 -88 -5) were used, which was considered suitable for read-across.

The key study was conducted with ethyl acrylate according to GLP and US EPA guideline OTS 797.1400 using rainbow trout (Salmo gairdneri, new name: Oncorhynchus mykiss) as test organism. Within this flow-through study, nominal test concentrations of 0, 0.44, 0.88, 1.8, 3.5, 7.0 and 14 mg/L were applied with corresponding mean measured concentrations of 0, 0.34, 0.78, 1.7, 3.3, 6.5 and 13 mg/L (recoveries between 77 and 95%, mean: 90 ± 6.6 %). As a result, a 96h-LC50 value of 4.6 mg/L was determined based on measured concentrations (arithmetic mean) (ABC Laboratories, 1990). Behavioral/sublethal effects noted during the study included on-bottom orientation, surfacing, dark discolorating, quiescence and loss of equilibrium (NOEC 0.78 mg/L).

Due to structural and property similarities with the tested analogue substance, it can be concluded that the above result also applies to 2-methoxyethyl acrylate (CAS 3121-61-7). For details see analogue justification in the overall summary of Chap 6.1 and in Chapter 13 of the Technical Dossier.
