Details on test conditions:
For the definitive study a series of test solutions were prepared by diluting a 1000 mg/L stock solution with ISO media to give final test media concentrations of 0, 3, 10, 30, 100 and 300 mg/L. A calculated aliquot of Pseudokirchneriella subcapitata inoculum was transferred into each test solution to give a nominal algal density of 10E4 cells/mL. Cell concentration was determined at 48 and 72 hours by haemocytometer. Test concentrations were determined every 24 hours by Liquid Chromatography/Mass Spectroscopy (LC/MS).

Blank flasks were prepared by removing an aliquot of test media from each volumetric flask before the Pseudokirchneriella subcapitata inoculum was added. Glassware was not conditioned prior to the start of the test. Test flasks were placed in illuminated orbital incubators according to the randomised distribution numbers generated by the Random.bas computer software. Incubator temperatures were recorded at 24-hour intervals for the duration of the test. Light intensity was not recorded.

A 10 mL aliquot of each solution was provided to Chemical Analysis at 0, 24, 48 and 72 hours for determination of test item concentration. Test concentrations were determined by LC/MS by the Chemical Analysis.

The algal concentrations in the test media were determined at 0, 48 and 72 hours using a haemocytometer.

Incubator temperatures ranged between 22 and 25 °C throughout the experimental period. The pH of each test solution was measured at 0 hours. The pH of the remaining test media in the first replicate flask of each test concentration was measured at 72 hours.

Details on results:
- Exponential growth in the control (for algal test): Yes, 110-fold increase in the cell density of control cultures over 72 h
- Observation of abnormalities (for algal test):
- No data
- Other obsevations:
The test item has limited solubility in water and ISO media. There are significant losses of the test item over a 72 hr period. At 0 hours the measured concentration of each chain length dissolved in solution is close to the expected nominal concentration taking into account the chain length distribution of the test item. The concentrations of C8, C10 and C12 remain stable over the test period however there is a significant decrease in the concentration of chain lengths ≥ C14 in solution. Comparisons between solutions with and without algae reveal a significant decrease in the total concentration remaining at 72 hours when algae is present (30% of the 100 mg/L solution remaining compared to 65% for the same concentration without algae).

Conclusions:
In this study the effects of the test item SLI(76) stripped on the growth of the freshwater green alga Pseudokirchneriella subcapitata were tested. The EC50-values for specific growth rate inhibition (ErC50) and inhibition of yield (EyC50) after 72 h were >= 1.87 mg/L (dissolved measured concentration). The NOEC values were >= 0.31 mg/L (dissolved measured concentration).
Executive summary:

This study was used to determine the 72-hour EC50 and NOEC (No Observed Effect Concentration) of SLI (76) stripped toPseudokirchneriella subcapitata.

SLI (76), provided by Unilever Mannheim was stripped of its fatty acid content via Soxhlet extraction at the Materials Science Laboratory, Trumbull, USA. The resulting material, SLI (76) stripped was used for this study.

Prior to testing the solubility of the test item and its individual actives were determined by Liquid Chromatography/Mass Spectroscopy (LC/MS) by the Chemical Analysis.

The test item has limited solubility in water and ISO media. There are significant losses of the test item over a 72 hr period. At 0 hours the measured concentration of each chain length dissolved in solution is close to the expected nominal concentration taking into account the chain length distribution of the test item. The concentrations of C8, C10and C12remain stable over the test period however there is a significant decrease in the concentration of chain lengths ≥ C14in solution. Comparisons between solutions with and without algae reveal a significant decrease in the total concentration remaining at 72 hours when algae is present (30% of the 100 mg/L solution remaining compared to 65% for the same concentration without algae).

For the definitive study a series of test solutions were preparedby diluting a 1000 mg/L stock solutionwith ISO media to give final test media concentrations of 0, 3, 10, 30, 100 and 300 mg/L. A calculated aliquot of Pseudokirchneriella subcapitata inoculum was transferred into each test solution to give a nominal algal density of 104cells/mL. Cell concentration was determined at 48 and 72 hours by haemocytometer. Test concentrations were determined every 24 hours by Liquid Chromatography/Mass Spectroscopy (LC/MS).

In this study the effects of the test item SLI(76) stripped on the growth of the freshwater green alga Pseudokirchneriella subcapitata were tested. The EC50-values for specific growth rate inhibition (ErC50) and inhibition of yield (EyC50) after 72 h were >= 1.87 mg/L (dissolved measured concentrations). The NOEC values were >= 0.31 mg/L (dissolved measured concentrations).

Description of key information

The toxicity of Fatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts to algae was tested according to OECD guideline 201. The ErC50 was observed to be >4.8 mg a. i. /L and the ErC10 was observed to be 0.3 mg a. i. /L

Key value for chemical safety assessment

EC50 for freshwater algae:
4.8 mg/L
EC10 or NOEC for freshwater algae:
0.3 mg/L

Additional information

There is noAlgal growth inhibition testavailable for lauric acid 2-sulfoethyl ester, sodium salt (sodium lauroyl isethionate) CAS No 7381-01-3.

There is however a study on the source chemicalFatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts (CAS no 85408-62-4), which isKlimisch rated 2and was carried out according to theOECD guideline 201under GLP,as described in Roberts, 2008 (EAL080070).

In this study the effects of Fatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts (CAS no 85408-62-4) on the growth of the freshwater green alga Pseudokirchneriella subcapitata were tested

The result of the algae test with Fatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts is read-across to coco fatty acids 2-sulfoethyl sodium salt. Read-across from the source chemical Fatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts (CAS no 85408-62-4) to the target chemical coco fatty acids 2-sulfoethyl sodium salt is considered to be justified as both substances are very similar and the source chemical contains a higher content of longer alkyl chain products which are known to be more toxic than the shorter alkyl chain products. The conclusion that the source chemical is more toxic than the target chemical is considered sufficient to select the more toxic derivative as the worst-case representative in the hazard assessment of both substances. For this reason read-across of ecotoxicity results of the source chemical (C12-18 and C18-unsatd) to the target chemical (Coco) is considered to be justified without the introduction of an additional safety factor.

Comparisons between solutions with and without algae reveal a significant decrease in the total concentration remaining at 72 hours when algae is present (30% of the 100 mg/L solution remaining compared to 65% for the same concentration without algae).

For the definitive study final test media concentrations were prepared of 0, 3, 10, 30, 100 and 300 mg/L. Cell concentration was determined at 48 and 72 hours. Test concentrations were determined every 24 hours by Liquid Chromatography/Mass Spectroscopy (LC/MS). The EC50-value for specific growth rate inhibition (ErC50) after 72 h was >= 4.87 mg/L (dissolved measured concentrations) based on active ingredient concentration. The NOEC value for specific growth rate inhibition was >= 0.31 mg/L (dissolved measured concentrations) based on test substance concentration and 0.3 mg/L based on active ingredient concentration.

Read-across from the source chemical, Fatty acids, C12-18 and C18-unsatd., 2-sulfoethyl esters, sodium salts to the target chemical is considered to be justified based upon a commonality of functional groups, constituents, breakdown products and metabolic pathways.

Therefore we consider these study endpoint results as sufficiently conservative to be taken account of in the risk assessment of lauric acid 2-sulfoethyl ester, sodium salt (sodium lauroyl isethionate) CAS No 7381-01-3.
