Skin irritation

1,5-Naphthylene diisocyanate (NDI) was slightly to moderately irritating to the skin of rabbits according to OECD TG 404 (Schuengel, 2006b). All signs were fully reversible within two weeks. No relevant systemic intolerance reactions were observed. In addition, a test using an artificial 3D-Skin model (OECD TG 431) revealed no corrosive properties of NDI in vitro (Vohr, 2006).

According to classification criteria NDI is not an irritant to the skin.

Eye irritation

NDI was slightly irritating to the eyes of rabbits according to OECD TG 405 (Schuengel, 2006c). In all animals redness of conjunctivae were fully reversible within three days. No systemic intolerance reactions were observed.

According to classification criteria NDI is not an irritant to the eyes.

Respiratory irritation

During acute inhalation testing NDI revealed concentration-dependent signs suggestive of irritation of the respiratory tract at concentrations of 96 mg/m3and higher (Pauluhn, 1995).

Justification for classification or non-classification

Skin irritation

Classified under Annex I of Directive 67/548/EEC with R38 (irritating to skin). This classification corresponds to Category 2 (H315: Causes skin irritation) according to Annex VI-1 of Regulation (EC) No 1272/2008. Differing from this no classification for skin irritation is required according to Annex I of Regulation (EC) No 1272/2008 based on the results of a dermal irritation/corrosion study on rabbits (OECD TG 404).

Eye irritation

Classified under Annex I of Directive 67/548/EEC with R36 (irritating to eye). This classification corresponds to Category 2 (H319: Causes serious eye irritation) according to Annex VI-1 of Regulation (EC) No 1272/2008. Differing from this no classification for eye irritation is required according to Annex I of Regulation (EC) No 1272/2008 based on the results of an eye irritation study on rabbits (OECD TG 405).

Respiratory irritation

Classified under Annex I of Directive 67/548/EEC with R37 (irritating to respiratory system). This classification corresponds to STOT-SE Category 3 (H335: May cause respiratory irritation) according to Annex VI-1 of Regulation (EC) No 1272/2008.
