Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Guideline:
other: Council Regulation (EC) No 440/2008, Method C.2 ‘Acute toxicity for Daphnia’ (2008)
Principles of method if other than guideline:
In most parts equivalent to OECD Guideline for Testing of Chemicals No. 202 'Daphnia sp., Acute Immobilisation Test' (adopted April 13, 2004)
GLP compliance:
yes (incl. QA statement)
Analytical monitoring:
yes
Vehicle:

Details on test solutions:
PRETREATMENT OF TEST ITEM:
- 105.4 mg of the test substance were added to 1 l of dilution water
- treated for 60 seconds with an ultra turrax, stirred for 1 hour on a  magnetic stirrer
- finally undissolved particles removed by filtration using using an aseptic filter (Serum Acrodisc® 37 mm Syringe Filter with GF/02 μm Supor® Membrane PN 4525 ).
Test organisms (species):
other: Daphnia magna STRAUS (Crustacea)
Details on test organisms:
- Test species: Parthenogenetic females, age 0 - 24 hours, laboratory bred at Currenta GmbH & Co. OHG, Leverkusen (Germany), cloned at the Federal Health Office (BGA), Berlin (Germany)
- Maintenance and acclimatisation: A population of parthenogenetic females of synchronized age structure has been maintained for more than 15 years in the test facility under constant temperature conditions (20 +/- 1 °C) at a 16 : 8 hour light-dark photoperiod (illumination: < 800 lux). The culture water (so-called 'M4 medium') was partly renewed once a week. The Daphnia were exclusively fed with unicellular green algae (Desmodesmus subspicatus) 'ad libitum'. Mortalities of parent Daphnia during the culture period were recorded daily in a semi-quantitative way. The neonates were separated from their parent Daphnia by filtration prior to the acute test.

Test type:

Details on test conditions:
TEST SYSTEM:
- keeping water: M4 medium, originally described in Water Research 24  (9), Sept. 1990; 1157-1167
- photoperiod: 16 hours light, 8 hours dark
- temperature of incubation unit : 19.6 to 19.9 °C
- feeding during test: none - test vessel: 50 ml glass beakers, 20 ml test medium, not ventilated
- number of daphnia per beaker: 10 daphnids (neonates)
- test parameter: immobility (death)
- method of administration: direct weighing
- test concentrations (nominal): 1 (100 mg/l) plus 1 control
- number of replicate: 2 (per concentration/control)
- medium renewal: none
Reference substance (positive control):
no
Duration:
48 h
Dose descriptor:
EC50
Effect conc.:
> 100 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Duration:
24 h
Dose descriptor:
EC50
Effect conc.:
> 100 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Details on results:
Recovery rates correspond to 99.9 % of nominal values at 0 hours, and to 94.9 % of nominal values at 48 hours, respectively.
Validity criteria fulfilled:
yes
Remarks:
The immobilisation and other abnormalities in the controls did not exceed 10 % by the end of the test. The dissolved oxygen concentration remained above 3 mg/L throughout the exposure period.
Conclusions:
For Daphnia magna STRAUS an EC 50 of > 100 mg/l was obtained after 48 hours.
Executive summary:

For Daphnia magna STRAUS an EC 50 of > 100 mg/l was obtained after 48 hours. The study was conducted according to Directive 92/69/EEC, C2.

The result relates to nominal concentration as analytical monitoring shows stability of the test substance during the experiment (Currenta, 2009).

Description of key information

For Daphnia magna STRAUS an EC 50 of > 100 mg/l was obtained after 48 hours. The study was conducted according to Directive 92/69/EEC, C2. The result relates to nominal concentration as analytical monitoring shows stability of the test substance during the experiment (Currenta, 2009).
