Description of key information

Intradermal and epidermal exposure to DAB concentrations (preliminary study for rangefinding for the sensitization study) induced necrosis, erythema and oedema. Some animals had to be sacrificed for ethical reasons shortly after exposure.
NOAEL: 2% (no erythema, oedema, nor sensitization)
Based on the data of skin corrosion, 1,4 Diaminobutane was judged to be corrosive to eyes as well.
Corrosive, R34 Causes burns
12 New Zealand White albino rabbits were exposed for 24 hours to 1,4 -diaminobutane (patch test); 6 animals on intact skin, 6 animals on abraded skin. After 48 and 72 hours the effects were evaluated, and scored according to Draize.
All animals had the maximum score on erythema (4), and this was not reversible during the observation period.
At the 24 hours observation period all animals had severe edema (3), wich was reverisble totally at the 72 hours observation period.
Average scores 24 hours: 6.8; 72 horus: 4.0.
CIVO scores: 24 hours: heamorrhages, ischemia and moderate edema; 72 hours: severe incrustation.
Exposure to 1,4 -butanediamine resulted in very severe skin irritation (corrosive).

Justification for classification or non-classification

Based on the observations in the dermal rabbit test and the dose range finding part of the Guineapig sensitization test 1,4 -diaminobutane is considered to be corrosive.

Exposure to 1,4 -butanediamine resulted in very severe skin irritation (corrosive).
