Additional information

Considering the acute toxicity to aquatic invertebrates, two key studies were identified, one for marine and one for freshwater invertebrates:
- To determine the toxicity of 1-Methyl-2-pyrrolidone to freshwater invertebrates, a static non-GLP study following German Industrial Standard DIN 38412 part 11 using Daphnia magna was performed (BASF AG, 1982). Based on nominal test concentrations, a 24 -h EC50 of >1000 mg/L was determined.
- As key study concerning acute toxicity to marine invertebrates a test performed by the Aquatic Toxicology Laboratory, SRC, Syracuse on behalf of GAF Corporation (1979) according to US EPA guideline 660/3 -75 -009 was identified. As test species the grass shrimp (Palaemonetes vulgaris) has been applied. In this static test, an EC50 (96h) of 1107 mg/L (nominal) was found.

According to REACH Guidance R.7b, section R.7.8.4.1 (ECHA, 2017) a test duration of 48 hours is standard for daphnids. 24 hour values should be considered only in the absence of good quality 48 hour values and in conjunction with other available data. For other crustacea, such as mysids or others, a duration of 96 hours is typical. Therefore, the available data (freshwater: 24-h EC50; marine: 96-h EC50) are sufficient in terms of chemical safety assessment.

Based on the available experimental data, 1-Methyl-2-pyrrolidone (CAS 872-50-4) is concluded to be with high probability acutely not harmful to aquatic invertebrates.
