Sodium perborate was tested positive without metabolic activation in bacterial reverse mutation assays with S. typhimurium and E. coli, in a DNA repair test with E. coli and in a test on chromosomal aberrations in CHO-K1 cells. With metabolic activation both cytotoxicity and mutagenicity were reduced. The data are indicating that this is maybe due to the generation of H2O2 (similar to investigations with H2O2 the responses observed were reduced by the presence of catalase). Therefore, this genotoxic potential may not be relevant in vivo. In addition, the uptake of sodium perborate into the cell is lower in comparison with H2O2.

In the EU RAR is was stated, that a.) the available studies are not in support of a significant genotoxicity/mutagenicity for hydrogen peroxide under in vivo conditions and b.) borates are negative in genotoxicity assays.
