Description of key information

A GLP-study in Oryzias latipes according to OECD guideline 204 is available for propylene glycol methyl ether acetate.

The Environmental Agency of Japan (1998) studied also the acute toxicity of propylene glycol methyl ether acetate to Oryzias latipes using flow-though conditions during 14 days. The experimental conditions were almost the same as the semi-static test. The exposure concentrations ranged from 40 to 100 mg/l and were measured by gas chromatography. Based on the geometric mean of the measured concentrations, the 14-day LC50 was determined to be 63.5 mg/l. The no observed effect concentration (NOEC) was determined to be 47.5 mg/l. The NOEC cannot be considered as a long term result and will only be used as supporting information. The test reports from the Environmental Agency of Japan (1998) are not available. Yet, all the data were validated by the Japanese authorities within the OECD SIDS program and will be taken into account for the determination of the PNEC for the aquatic compartment.

Propylene glycol methyl ether acetate is classified as readily biodegradable. The risk assessment shows that the PEC/PNEC ratio is clearly < 1 in surface water, indicating no need for further information or testing. According to REACH Annex IX requirements, long-term toxicity testing shall only be considered when the chemical safety assessment indicates the need for further investigations. Hence, further long-term testing in fish can be waived for propylene glycol methyl ether acetate.
