2 -ethoxyethanol was tested for 48 h at various concentrations between 0.1 M and 0.1 µM, using serial half-dilutions. Dose-response curves showed the common fearture of a very steep slope on a semi-log plot, indicating that cell viability went from normal to nil by increasing concentration. This steep slope and the sigmoid shape of the curve suggested that the substance hindered some cellular mechanisms essential for cell survival.

IC50 for 48 h incubation corresponded to the concentration (mM) for which the cell viability in treated culture were 50 % of the viability in untreated control cultures.

Conclusions:
The test substance 2-ethoxyethanol was found to be a hemopoietic toxin and acted directly on the hemopoietic cells, either growth-factor-dependent or leukemic from rat, mouse and human species.
