Approximately 0.08 g of test substance was placed in a 20 mL LSC vial and brought to volume with mobile phase (55% methanol:45% water). A calibration curve using structurally related reference compounds of known Log Koc was established, using methanol or mobile phase. The test substance and reference compounds of known Log Koc. were injected into an HPLC system. A calibration curve using retention time (Log t) versus Logo Koc. was calculated by linear regression analysis from the injection data of the reference compounds. The calibration curve was calculated from literature values for the Log Koc of the reference compounds and the corresponding capacity factor, k, calculated as:

K = (tr-to) / to where tr is the retention time of the reference compound

From this curve, the unknown Log Koc of the test substance was estimated from its retention time by making injections of small samples of the test substance following identical column, elution, and detection parameters used to establish the calibration curve. The Log Koc of the test substance was determined by calculating the capacity factor (k) of the test substance.

The HPLC system used for the analysis consisted of: Varian Prostar HPLC with Varian Model 410 autosampler equipped with a partial loop injector. Data capture was by Galaxie System software. The colunm was Phenomenex Luna C18 with 5 jam particle size x 250 mm length x 4.6 mm inner diameter; Phenomenex Cyano 5 um particle size x 250 mm length x 4.6 mm inner diameter for Log Koc.

The HPLC operational parameters were as follows: Detector; PDA, Flow rate (mobile phase); 0.8 mL/min. Injection volume; 10 1ul, Detector wavelength; 210 nm, Colunm temperature; Room Temperature, Attenuation / Chart Speed; Autoscale, Threshold / Peak Width 1000 / 2.0 and Mobile phase solvent; (55:45 ratio of methanol/water).

The Log Koc of the test substance was determined to be 2.32 and the adsorption coefficient (Koc) was approximately 210 when estimated by comparing the retention times of the multiple reference compounds of known Log Koc. eluted through a reverse phase HPLC column.
